1) It seems more natural to me to say,
- He looks very much like you.

2) I have one brother. - emphasizes the number.

- I have a brother. - You could well use this if you have one brother, but you would not be putting the same emphasis on the number one.

You might also use "a brother" if you are going to talk about a particular brother but you actually have more than one.
